Weghorst, who was born in Borne, Overijssel, began his career with local clubs RKSV NEO and DETO Twenterand.

He later joined Willem II of the Eredivisie. Despite having the opportunity to break into the first team, he could not do so and so signed for Eerste Divisie club FC Emmen.

On August 10, 2012, he debuted in a match against Dordrecht, which ended 1–1.

Weghorst would score his first goal in professional football a month later, in the derby against Veendam, which Emmen won 2–1.

He appeared in 28 games and scored eight goals in his first season. In addition, Weghorst signed a free transfer contract with Eredivisie team Heracles Almelo after his time with Emmen.

Furthermore, Weghorst made his Eredivisie debut for Heracles Almelo on August 9th, in a 0–3 home loss against AZ Alkmaar at the Polman Stadion.

During his two years in Almelo, he was a consistent starter. Unfortunately, the club was relegated from the Eredivisie in his first season, but he had a more successful second season with Heracles.

Weghorst signed a four-year contract with VfL Wolfsburg on June 26, 2018.

In a 5–2 league win over Fortuna Düsseldorf on March 16, 2019, he scored his first hat-trick for Wolfsburg and the first by any player for the club since Mario Gómez in April 2017.

He scored 17 goals in his debut season in Germany, placing him joint-third in the Bundesliga goalscoring lists for the 2018–19 season.

Weghorst had another outstanding goal return in the 2019–20 season, scoring 16 times in the league as Wolfsburg placed sixth.
